Grant Detail | The objective of this study is to determine the effects of maternal obesity and diabetes on infantile body composition, growth and epigenetic changes related to free fatty acid/glucose metabolism and diabetes. Body composition is measured serially utilizing the air displacement plethysmography and dual-enegry X-ray absorptiometry. Cord blood is collected for analyses of promoter methylation status of key genes in the insulin signaling, glucose metabolism and free fatty acid pathways. |

Grant Detail | The primary goal of this project is to investigate the effect of hyperglycemia on pancreatic development and peripheral insulin signaling at critical periods of development in the premature baboon. The central hypothesis is that preterm baboons have decreased pancreatic function and further impairments of insulin signaling in skeletal muscle when compared with preterm baboons not exposed to adverse postnatal environment (hyperglycemia). Pancreatic function is assessed by hyperglycemic clamps and compared to histological endocrine cell development utilizing the CAST system. Insulin signaling molecules and glucose transporters are measured at basal and insulin-stimulated state in skeletal muscle. |

Grant Detail | The primary goal of this project is to determine if differentially methylated regions in cord blood of infants of mothers with gestational diabetes and obesity are associated with increased adiposity in offspring. Body composition is measured serially (birth, 3 months and 1 year) by utilizing the air displacement plethysmography and dual-energy x-ray absorptiometry. Methylation status of key genes in the insulin signaling, glucose metabolism, and free fatty acid pathways are measured at 1 year of age. |
